The settlement emerged in 1900 during the construction of the Moscow-Ventspils railway line, originally centered around a station known as Rosenau. Its strategic location as a border gateway led to rapid development, and it was granted town rights in 1931. The town's infrastructure is heavily influenced by its status as a major international rail transit point and border crossing. Culturally, Zilupe is diverse, housing both Russian Orthodox and Roman Catholic congregations.
